Effect of Estrogen on Hemopoietic Organs of Mice



Abstract
The present experiment was designed to.investigate the effects of large dose of estrogen on the changes of peripheral blood and the histopathological changes of extramedullary hemopoietic organs, namely liver and spleen of mice.
Histopathological changes and weights of liver and spleen, and peripheral blood counts of erythrocytes and leukocytes were observed after subcustaneous injection of 25, 50, and 100 fig per mouse of estradiol-benzoate for one, two, and four weeks, respectively.
The following results were obtained.
1. The nucleated erythroid i recursors were found on the wall of hepatic vein and portal areas of liver after treatment for four weeks in all experimental mice, however these findings were disappeared by four weeks after cessation of treatment. In the spleen, the nucleated erythroid precursors were scattered around the splenic sinus.
2. The weight of liver was increased after treatment for four weeks in all experimental mice, and returned to normal range by four weeks after cessation of treatment. But the weight of spleen . was increased after treatment for two weeks, and persisted in increased -state by four weeks after. cessation of treatment.
3. There was no significant difference between control and experimental group in the peripheral blood counts of erythrocytes. However, in leukocytes counts, leukopenia was observed after treatment¢¥.for two weeks and was more and more prominent with increasing the treated dosage. After cessation of treatment, the leukocyte count was increased gradually and reached to normal range by four weeks after cessation of treatment. And the degree of leukopenia was also rriore prominent in large dosage treated group.
